PCI Express Slot x16 question?

Ok, i have an asus mobo with 2 x16 express slots. I also have a giant heatsink and fan over my cpu, so it is getting a little close to the graphics card, I am wondering if I can move the graphics card down to the second x16 slot, or will it only work if there are two graphics cards

It should run just fine in the second slot providing you have the correct info on the slot. I had to do that once and it worked on my Asus board.
You may have to change the display in the BIOS but it usually does it automatically.
